PX-12 inhibits TG2 in the intestinal lamina propria. (A) Unfixed cryosections (400 nm) of duodenal biopsies were incubated with 5BP in the presence of increasing concentrations of DTT. Increasing amounts of DTT activate TG2 in the lamina propria. (B) TG2-activity (green) was normalized against TG2-expression (red) and quantified using ImageJ. Higher amounts of DTT led to increased TG2 activity. n = 6; 5 images per patient were quantified. (CF) Cryosections were treated with ERW1041 and PX-12 prior to the incubation with 5BP. Both inhibitors significantly reduced TG2 activity in the lamina propria. n = 6; 5 images per patient were quantified. All data are shown as mean ± SD.
