Lactate is incorporated in cellular metabolism and displaces glucose. (A) Experimental design. Mouse CD8+ T cells were activated in the presence of either 11.1 mM [U-13C6]glucose + 40 mM NaCl, or 11.1 mM [U-13C6]glucose +40 mM sodium lactate, or 11.1 mM glucose and 40 mM [U-13C6]sodium lactate. Incorporation of labelled carbons in cellular metabolites was assessed by GC-MS at 24, 48, and 72 hours after activation.
