The HeLa cell lines with reduced levels of all sHSPs and severe mitochondrial defects (used in Fig. 3) were generated by the combination of CRISPR/Cas9 knockouts and short hairpin RNAs (shRNA). Knockout or knockdown of individual components (HSPB1, HSPB5, HSPB8) did not present alterations in mitochondrial morphology compared to wild-type HeLa cells.
