Upon this binding, the SMO's inhibitory block is removed, which activates a group of GLI proteins (the Hh signaling pathway transcription factors). In response to activation of the Hh signal, GLI proteins are differentially phosphorylated and rebuilt into transcriptional activators that activate the expression of Hh target genes by direct interaction with specific regions in the promoter region
