Graphical abstract: The dual role of ADAM17 on EphA2 signaling. Based on our data, ADAM17 influences EphA2 signaling by two mechanisms: 1) Release of growth factors that act on EphA2 pS897 via the MAPK pathway and thereby activate the non-canonical pathway. 2) Cleavage of GPI-anchored ephrin-A1 to prevent receptor internalization and thereby blocking the canonical signaling pathway.
