IMD-0354 reverses nuclear translocation of NF-κB-p65 in microglia in a high glucose environment. (A-F) Western blot analysis showed that high glucose promoted NF-κB-p65 expression and its nuclear translocation in microglia. Inhibition of IKK2 by IMD-0354 downregulated the expression of NF-κB-p65 and blocked its nuclear translocation (E) under high glucose conditions, whereas the cytoplasmic levels of NF-κB-p65 were unaffected (F). Data are shown as the mean ± SD (****, P<0.0001; ***/###, P<0.001; ##, P<0.01). HG, high glucose; NF-κB-p65, nuclear factor κB-p65; GAPDH, glyceraldehyde-3-phosphate dehydrogenase; SD, standard deviation.
