IMD-0354 reverses nuclear translocation of NF-κB-p65 in Müller cells in a high glucose environment. (A-F) Western blot analysis showed that high glucose promoted NF-κB-p65 expression and its nuclear translocation towards in Müller cells. IKK2 by IMD-0354 downregulated the expression of NF-κB-p65 and blocked its nuclear translocation (E) in response to high glucose, whereas the cytoplasmic levels of NF-κB-p65 were unaffected (F). Data are shown as the mean ± SD (****, P<0.0001; ***, P<0.001; ##/**, P<0.01; #, P<0.05). HG, high glucose; NF-κB-p65, nuclear factor κB-p65; IKK2, inhibition of IκB kinases 2; SD, standard deviation.
