MMP inhibitory activity of N-TIMP2 variants. MMP-2_ACT (A,B), MMP-9_CAT (C,D) and MMP-14_CAT (E,F) were incubated with N-TIMP2, N-TIMP2-DOPA variants (A,C,E) or N-TIMP2-HqAla (B,D,F) variants at various concentrations. Cleavage of the chromogenic substrate (100 µM) was measured over time, and the velocity (slope) of the reaction as a function of inhibitor concentration was fitted to Morrison's equation (Eq. 1) to obtain the inhibition constant K_i. Statistical analysis for the comparison of N-TIMP2 to N-TIMP2-DOPA and N-TIMP2-HqAla variants was performed by Student's t-test; *P < 0.05, **P < 0.01. Error bars represent SEM; n = 3.
