Multiple functions of the proteasome in the nervous system. The proteasome plays critical roles in a wide range of functions critical to the nervous system. These include protein degradation, both of oxidized and misfolded proteins as well as degradation of polyubiquitinated proteins. The proteasome impacts a range of functions specific to the nervous system, including synaptic physiology and plasticity, neurodevelopment and glial cell function, and maintenance of circadian rhythm. The proteasome modulates learning and memory. Proteasome impairment is a robust feature both of aging and neurodegenerative disease, leading to dysfunction in these pathways.
