Mechanism of action of tecovirimat, brincidofovir and cidofovir. The diagram depicts a probable lifecycle of the mpox virus. Tecovirimat hinders viral envelope formation of orthopoxviruses (e.g., mpox and smallpox) by inhibiting p37 protein, a highly conserved sequence among all orthopoxviruses. Brincidofovir, a prodrug and lipid conjugate of cidofovir, can easily pass through the phospholipid bilayer compared with cidofovir.
