Fig. 6 — Mechanism of action of tecovirimat, brincidofovir and cidofovir. The diagram depicts a probable lifecycle of the mpox virus. Tecovirimat hinders viral envelope formation of orthopoxviruses (e.g., mpox and smallpox) by inhibiting p37 protein, a highly conserved sequence among all orthopoxviruses. Brincidofovir, a prodrug and lipid conjugate of cidofovir, can easily pass through the phospholipid bilayer compared with cidofovir. Cidofovir diphosphate competitively inhibits the viral DNA polymerase and thus hinders viral DNA replication. mpox monkey pox
