(A, B) Graphs illustrate a quantification of CD122 and CD132 expressions (geometric mean) on NK cells (NK1.1+ CD3/19) of the spleen (A) and BM (B) from mice from the indicated genotypes. (C) Total NK cells were enriched from the spleens of Ncr1cre Mycfl/fl and Ncr1cre Mycwt/wt mice and the protein expression levels of STAT5, Erk2, S6, and Rpl11 were assessed by Western blotting. Histone H3 levels are shown as loading controls. (D) DERL-7 cells were pretreated (30 min) or not with Torin2 (250 nM), PD98059 (10 μM), tofacitinib (90 nM), and then stimulated with 25 ng/ml IL-15 for 35 min and the phosphorylation of STAT5, Erk, S6, and p70 S6 kinase was assessed by Western blot. β-Actin levels are shown as loading control.
