Table 1.
Optimum method parameters determined for the flow-cytometry-based quantification of BVs after the infection of Sf9 cells in a 96-well format. The procedure without staining is only suitable for viruses expressing a fluorophore (e.g., GFP), whereas the staining procedure targets the gp64 surface protein, which is present on all BVs, independent of the genotype.
| Parameter [Unit] | No Staining | PE Staining | APC Staining |
|---|---|---|---|
| Seeding cell concentration [cells/mL] | 0.8 × 106 | 1.0 × 106 | 1.0 × 106 |
| Volume per well [µL] | 150 | 150 | 150 |
| Incubation time of virus infection [h] | 17 | 5 | 4 |
| Concentration of fluorescent antibody [mg/mL] | Not applicable | 0.001 | 0.00025 |
| Volume per well [µL] | 50 | 50 | |
| Incubation time of antibody staining [min] | Not applicable | 30 | 15 * |
* The incubation can be performed at 4 °C or at room temperature, without affecting the results.
