(A) Illustration of DepMap data (Meyers et al., 2017 and Dempster et al., 2019) focusing on expression and dependency score (CERES) of TP73 in 34 PDAC cell lines. CERES score less than −0.5 considered as dependent, and the two dependent cell lines, KP2 and HS766T, are labeled. (B) Validation of TP73 dependency using competition-based cell proliferation assays in TP73-dependent (KP2 and HS766T) and TP73-neutral (PATU8902 and MIAPACA2) cell lines. Cas9-expressing cell lines were infected with sgRNAs whose expression plasmids also encode GFP. Non-targeting sgRNAs (N), sgRNA targeting PCNA (positive control), sgRNA targeting all isoforms of p73 (sgRNA #1 and #2, red), and sgRNAs targeting p73-TA (sgRNA #1 and #2, green) were used. Percentage of GFP+ cells was measured every 2 or 3 days (P1-P7) starting day 3 (P0) post-sgRNA infection. For each sgRNA experiment, every data point is normalized to the percent GFP value at P0.
