Determination of the minimal effective MOI (mMOINF-κB) of commensal bacteria based on NF-κB pathway activation in (3D) A549 luciferase NF-κB-reporter cells. Selected lung commensals at different MOI were cultured with LPS and the mMOINF-κB was determined by a NF-κB luciferase assay, with (A) A. naeslundii, (B) G. haemolysans, (C) G. sanguinis, (D) M. luteus, (E) P. histicola, (F) P. melaninogenica, (G) P. nigrescens, (H) R. mucilaginosa, (I) S. sanguinis. On the vertical axis % NF-κB pathway activation compared to LPS-stimulated cells is shown, on the horizontal axis the tested MOI is depicted. The mMOINF-κB was defined as the lowest MOI with anti-inflammatory activity (i.e.< 50% NF-κB pathway activation compared to LPS-stimulated cells as visualized by a green line, p< 0,05). *: p< 0.05, n ≥ 3.
