Accumulation of SafA and SpoVID in sporulating cells and mature spores. Whole-cell extracts were prepared from the wild type or mutant strains at the indicated times (in hours) after the onset of sporulation. Spore coat extracts were prepared from mature spores as described in Materials and Methods. These extracts were subjected to electrophoresis in SDS-polyacrylamide gels, and immunoblots of the gels were stained with anti-SafA (A) or anti-SpoVID (B) antiserum. Extracts from the wild-type strain were harvested 2, 4, 6, 8, and 30 h after the onset of sporulation as indicated above the lanes, but extracts from safA, spoVID, and sigE mutants were harvested 4 h after the onset of sporulation. Arrows A and B indicate the 43- and 30-kDa species of SafA, respectively.
