Determination of apparent K_m values for orotate uptake. S. meliloti Rm8002 cultures were grown in M9 medium with 0.2% aspartate as the carbon source. The cultures were washed and resuspended in M9 medium without a carbon source. Assays were performed using [³H]orotate. The uptake rate (V) at different concentrations of orotate (S) is plotted. The insert shows the Eadie-Hofstee plot of V/S versus V that was used to calculate an apparent K_m value.
