Schematic of experimental design. Three-month-old pigs were fed either standard pig chow (Lean) or a high-fat/high-fructose diet (Obese) for 16 weeks (n = 6 each). Mesenchymal stem/stromal cells (MSCs) were isolated from subcutaneous abdominal fat, expanded in vitro, and characterized for surface markers. Hydroxymethylated DNA immunoprecipitation sequencing (hMeDIP-seq) was performed on Lean- and Obese-MSCs (n = 3 each), and again on Obese-MSCs (n = 3) cultured with epigenetic modulator vitamin-C, to determine levels of the 5-hydroxymethylcytosine (5hmC) epigenetic mark.
