Chromosome Walking toward the RSF1 Gene.
(A) Genetic map positions, in relation to other known phenotypic and molecular markers, of SGCSNP5 and SGCSNP247, the two SNPs that mapped the rsf1 mutation to the top of chromosome 1 by array hybridization.
(B) Physical map of the BACs sequenced between SGCSNP5 and SGCSNP247 as well as the positions of the seven markers (see Table 1) used to shorten the interval by DHPLC-based genotyping of 692 F2 plants. The 14 recombinants that were heterozygous at SGCSNP5 were gradually reduced to one recombinant at T7I23.96000. Correspondingly, the 17 recombinants heterozygous at SGCSNP247 were reduced to three at T14P4.5700. The remaining gap between markers T7I23.96000 and T14P4.5700 occupied 55,331 bp.
