Comparison of tau mutants reveal that within the 0N3R isoform, S356T is uniquely capable of K19 seed-induced aggregation. HEK293T cells were transfected to express either (A) WT tau or different tau mutations (B) G335V, (C) S352L, (D) S356T, (E) V363A, (F) V363I, (G), and G366R and untreated or treated with K19 seeds. Biochemical aggregation assays were performed as described in Materials and Methods and assessed by immunoblotting with tau antibody 3026. The relative mobilities of molecular weight markers are shown on the left. (H) Graph shows percent aggregation of 0N3R WT tau and different tau mutations.
