Overview of using biophysical computational models to evaluate causal interventions. Empirical structural connectivity between regions can be reconstructed from diffusion MRI tractography, which provides the global connectivity between regions. Perturbations can be applied to this connectivity, to simulate lesions or plasticity. Additionally, perturbations to the local dynamics of each region can simulate neuromodulatory influences and pharmacology. The resulting simulated activity and functional connectivity can then be compared against empirical data, to evaluate the model's goodness-of-fit.
