Development of the model of innate immune memory in non-immune human osteoblast-like MG-63. Human osteoblast-like MG-63 cells were seeded with an initial concentration of 10,000 cells/well, 20,000 cell/well or 30,000 cell/well into 24 well-plates in the medium containing either 2.5%, 5%, 7% or 10% of FCS. The number of cells at the end of the experiment (7th day) was determined by a Cell counter (Bio-Rad) using samples prepared for each concentration of FCS corresponding to the different initial cell concentrations. Data are presented as mean +/- SD. Each experiment was done in triplicate. The differences among the groups corresponding to different conditions were assessed by analysis of variance (ANOVA) followed by a Tukey’s Honestly Significant Difference test. (*) P-values, ≤ 0.05 and (***) P-values ≤ 0.005 were considered to be significant.
