Spectrally determined K_dvalues of P450 51A1–sterol complexes. Two cuvettes containing 1.0 μM P450 51A1 were placed in a spectrophotometer, and the indicated concentrations of the individual compounds were added to the sample cuvette. The insets show spectral traces used in each analysis. The data were fit to a hyperbolic curve, using a quadratic equation due to the low K_d values. A, dihydrolanosterol (K_d 0.23 ± 0.10 μM); B, 14α-CH₂OH dihydrolanosterol (K_d 0.02 ± 0.04 μM); C, 14α-CHO dihydrolanosterol (K_d 0.12 ± 0.03 μM); D, dihydro FF-MAS (K_d 1.4 ± 0.5 μM). FF-MAS, follicular fluid meiosis-activating sterol ((4β,5α)-4,4-dimethyl-cholesta-8,14,24-trien-3-ol).
