Detection of CysS-(S)n-H in CSEs. Purified wild-type (WT) or indicated mutants were incubated with 1 mM cystine or buffer alone in 50 mM HEPES (pH 7.5) and 50 µM PLP. Polysulfidated CSE was detected by a biotin-PEG-MAL capture assay. The histogram shows the amounts of polysulfidated CSE relative to those of expressed CSE (Total CSE). Results are the mean ± SE of five to six independent experiments. *** p < 0.001 and ** p < 0.01 when compared with control of each sample without cystine. n.s.: not significant.
