Old mice (14–17 m.o. at priming) were immunized IN with 10 μg RBD with PBS, NE, or NE/IVT, or IM with RBD with Addavax. Controls included mice immunized IN with PBS or NE/IVT without antigen. Mice were given three immunizations at a 3-week interval. Serum (a–c) RBD- and d–f S protein-specific IgG was measured 2 weeks after (a, d) prime (b, e) prime/boost and c, f prime/boost/boost immunizations. Neutralizing antibody titers were measured in sera after (g–j) prime/boost and d–n prime/boost/boost immunizations with SARS-CoV-2 PSVs for (g, k) WT, h, l B.1.617.2, i, m B.1.351, j, n B.1.1.529 variants. Titers are shown as mean ± SEM (n = 5/grp) (*p < 0.05, **p < 0.01 by Mann–Whitney U test shown between adjuvanted groups).
