Western blot analysis of lysates from MDA-MB-231 cells treated with 25 or 50 μmol/L of Ac₄ManNTz (1), Ac₄ManNPhTz (2), Ac₄ManNPh (3) or DMSO (no tetrazine-sugar). MDA-MB-231 cells were incubated with biotin-TCO probe (0.25 mmol/L) for 30 min. The cell lysates were harvested and probed with anti-biotin antibody conjugated to horseradish peroxidase (HRP) (upper panel), then stripped and re-probed with an anti-GAPDH antibody as a protein loading control (lower panel).
