(A) CD45 was used to identify all hematopoietic cells, and macrophages were considered CD45 CD11b double positive. Other immune cells, including CD3+ T cells, CD15+ neutrophils, Siglec8+ eosinophils were excluded. After identifying monocytes (CD45+FSClowSSClow) using forward and side scatter, CD14 and CD16 expression was used to identify non-classical, classical, and intermediate subtypes. (B) Macrophages were further categorized into M1 or M2 using CD86 and CD206 respectively, as well as pro-inflammatory marker S100A9, and tissue resident marker CD169.
