Vav1 knockdown represses inflammation in the ischemic penumbra of MCAO/R rats.

(A, B) NLRP3, ASC, and cleaved caspase-1 (p20) expression levels in the ischemic penumbra area in MCAO/R rats (normalized to the sham group) were analyzed by western blot. (C) IL-1β, TNF-α, and IL-18 mRNA expression levels (normalized to the sham group) in the ischemic penumbra area of MCAO/R rats were analyzed by qPCR. (D) The contents of IL-1β, TNF-α, and IL-18 in the ischemic penumbra area of MCAO/R rats were analyzed by ELISA. Data are expressed as the mean ± SD (n = 6), and were analyzed by one-way analysis of variance followed by Tukey’s multiple comparisons test. The experiments were repeated three times. ASC: Apoptosis-associated speck-like protein; ELISA: enzyme-linked immunosorbent assay; IL-18: interleukin-18; IL-1β: interleukin-1β; MCAO/R: middle cerebral artery occlusion/reperfusion; NLRP3: NOD-like receptor pyrin 3; qPCR: quantitative polymerase chain reaction; TNF-α: tumor necrosis factor-α; Vav1: Vav guanine nucleotide exchange factor 1.
