The molecular network of Sirt6 in cardiac ischemia/reperfusion (I/R) injury. Sirt6 activation disassembles CHMP2B accumulation via modulation of FoxO1-Atrogin1 signaling pathway and attenuates cardiac I/R injury. Sirt6 reduces myocardial susceptibility to I/R insult through preserving mitochondrial quality control via SIRT6-AMPK-PGC1α-AKT signaling pathway. Sirt6 inhibits apoptosis via NF-κB signaling. Sirt6 interacts with MiR-144 to reduce H/R-induced myocardial injury. Sirt6/FoxO3 complex upregulates the transcriptional activity of FoxO-dependent antioxidant genes to counteract I/R insult.
