Figure 1: Schematic of the microfabricated multi-well devices.
(A) Individual C. elegans are cultured on solid low-melt nematode growth media (lmNGM) agarose pads seeded with bacterial food in individual wells. The space between the wells is coated with an aversive chemical (copper sulfate) to isolate each worm within its well. Each device is secured inside a single-well tray. The perimeter of the tray is filled with water crystals to maintain the humidity. The tray is sealed with Parafilm to allow oxygen exchange. Image created with BioRender.com. (B) Overview of the multi-well device indicating the suggested order for loading the wells. The inner wells (white) receive 14 μL of lmNGM. The outer wells (gray) receive 15 μL of lmNGM.
