Amplification time optimization for the HIV-1-RT-LAMP-AuNPs-LFA assay. Four RT-LAMP reaction times were evaluated at 68°C: (A) 15 min, (B) 25 min, (C) 35 min, and (D) 45 min. Tube/biosensor 1-7 represent nucleic acid template levels 2.0 × 10⁴, 2.0 × 10³, 2.0 × 10², 2.0 × 10¹, 2.0 × 10⁰, and 2.0 × 10⁻¹ copies and blank control (DW), respectively. Results were analyzed using visual reagent L-HNB and AuNPs-LFA. The optimal LoD occurred with a 35 min reaction time. DW, distilled water; CL, control line; TL, test line.
