Melibiose-stimulated proton transport. Melibiose was added to a final concentration of 10 mM, and the change in extracellular pH was monitored with a pH electrode and chart recorder. A downward deflection indicates alkalinization of the extracellular medium caused by sugar-stimulated proton uptake into the cell. The R52S, R52V, and R52Q mutant strains gave similar results in this experiment and are all represented by a single tracing.
