Synergistic antitumour activity of irradiation and RIG-I activation in vivo depends on functional p53 in melanoma. (A) Wildtype or p53/ B16 cells were subcutaneously transplanted into C57/BL6 mice. The mice were then locally irradiated (2 Gy), injected with 20 µg 3pRNA, or treated by both. After 16 h, the mice were sacrificed. Tumour-draining lymph nodes were analyzed by flow cytometry for CD69 surface expression of activated CD8+ T cells, CD4+ T cells, and NK1.1+ NK cells. Mean ± SEM of n = 3 with 3–5 mice per group and experiment. (B) Mice were treated as indicated over 7 days and the tumour size was measured daily.
