The variant HIV-1 enhancer binds a new nuclear protein. EMSA was performed with HeLa nuclear extract (n.e.) stimulated (+) or unstimulated (−) with TNF-α (lanes 1 to 12) and probes wt-wt and mut-wt as indicated above the lanes. In lanes 13 and 14, purified recombinant NF-κB p50 was used for EMSA. Monoclonal antibodies specific for the NF-κB subunits RelA (lanes 9 to 12) and p50 (lanes 5 to 8) were used to supershift the NF-κB complex. Positions of the NF-κB, p50, and supershift complexes are indicated.
