PR-619 inhibited cell proliferation and induced cell cycle arrest at G0/G1 in chondrosarcoma cells. JJ012 and SW1353 cells were treated with 5 μM PR-619 and DMSO (for the untreated control) for 48 h. A. Cell proliferation was analyzed using the BrdU assay. B. Flow cytometric analyses were used for cell cycle progression and quantitative data are presented as the means ± SD from at least three independent experiments; *P<0.05 was considered significant as compared with the control. C. JJ012 and SW1353 cells were treated with 2.5 and 5 μM PR-619, respectively, with DMSO for the nontreated control, for 48 h. Cell lysates were harvested for western blot analyses with specific antibodies for phospho-AKT, AKT, phospho-ERK, ERK, p21, phospho-histone H3, and PCNA; all results are representative of at least three independent experiments.
