(B) The enzymatic data in (A) were fitted to Michaelis-Menten kinetics. Vmax and Km were calculated for different JSD26 concentrations (n=3). (C) Differential scanning fluorimetry (DSF)/thermal shift assay probing protein stabilization of purified SCoR2 by JSD26 and NADPH (substrate). The rate of change in SYPRO orange fluorescence is plotted versus temperature for SCoR2 in the absence and presence of combinations of NADPH, NADP+, and JSD26. Experiments were done in duplicate.
