AMIGO2 inhibited GSDME‐mediated pyroptosis through (caspase‐8 and caspase‐9)/caspase‐3 cascade. A549/shAMIGO2 cells pretreated with Av‐DEVD‐CHO (100 μM), Z‐LEHD‐FMK (50 μM) or Z‐IETD‐FMK (50 μM) for 4 h were exposed to 5 μg/mL cisplatin for 24 h. (A) MTT method was performed to assess cell viability. (B) Cells were lysed to obtain protein samples for western blot analysis with the indicated antibodies. (C) Cells were stained with PI and Hoechst 33342 prior to capturing the fluorescent images (20× objective lens). Five fields were randomly selected to calculate PI‐positive cells for statistical analysis.
