Healthy donor PBMC were isolated from buffy coat and rested overnight in 10 ng/mL IL-15. NK cell subsets were flow sorted and co-cultured with K562 WT and all-stimulator B cells at 1:1 E/T for 5 hours to assess NK cell killing and activation. a K562 WT cell death and b allo-stimulator cell death were increased by addition of NK cells in culture. Error bars show standard deviation of mean on technical replicates. c Ksp37 release by NK cells was positively correlated with percent of K562 WT and d allo-stimulator cell death. High cell death and Ksp37 release was dominated by educated NK cell subsets and high CD107a expression. P value in a and b was calculated using two-way ANOVA as indicated, and as 2-sided unpaired Student’s t-test adjusted by Tukey’s multiple comparisons correction. P value in c and d are calculated by Pearson’s correlation.
