Demonstration of the impact on the relationship between external concentration (mg/kg bw/day) and area under the curve (AUC) of plasma concentrations during the last 24 hours (μmole*h/L; AUC) from decreasing or increasing the values of the maximum rate of hepatic metabolism of the parent (Vmax) by 2, 5, and 10-fold (panel A); decreasing or increasing the Michaelis-Menten constant (Km) by 5, 10, and 100-fold (panel B); decreasing or increasing Vmax and Km together by 10-fold (panel C); or setting the in relative oral absorption fraction (fimax) and dose associated with half-maximal absorption (f50) to (0.9, 100), (0.5, 100), (0.1, 100), (0.9, 500) and (0.9, 50) (panel D). The results are presented for concentrations above 10 mg/kg bw/day to emphasize the divergent results produced by changing fimax and f50.
