Table 1.

Rates of transitions between the NR and R conformations.

Buffer FRET pair Experimental condition kR→NR, S−1 kR→NR, S−1
Polyamine buffer S6-Cy5/L9-Cy3 70S No antibiotic 0.3 0.5
Blasticidin S 2 0.2

Blasticidin S 2.2 0.3
Polymix buffer S6-Cy5/L9-Cy3 70S No antibiotic 0.7 0.8
Blasticidin S 0.9 0.3
h44-Cy3/H101-Cy5 70S No antibiotic nd nd
Blasticidin S 0.8 0.6

Rates of transitions between the NR and R conformations in S6-Cy5/L9-Cy3 and h44-Cy3/H101-Cy5 ribosomes bound with P-site deacylated tRNAfMet that were imaged in absence of antibiotics or in presence of BlaS. Rates were determined by fitting dwell time distributions (1 second binning size) to single exponential decay function. nd – not determined due lack of fluctuations.
