WT (CD45.1/1) and KO (CD45.1/2) OT1 cells were co-transferred into CD45.2/2 C57BL/6 recipients. One day later, the mice were vaccinated with OVA-encoding mRNA-LNPs as described in the STAR Methods. On day 10 post vaccination, the spleens were harvested, and the WT:KO ratio and phenotype of the responding OT1s were was analyzed by flow cytometry.
