Bifunctional degraders simultaneously bind to the protein of interest (POI) and LTR, forming a ternary complex. This receptor-ligand interaction initiates the internalization of the entire complex via endocytosis pathways. Once inside the endosome, the LTR dissociates from its ligand and recycles back to the cell surface, while the degrader and a part of the POI are likely directed to the lysosome for degradation afterward.
