(A-E) PEO1-OR cells were transduced with the indicated targeted shRNA or a non-targeting scrambled shRNA (shCTRL) and then selected in puromycin. For each knockdown or control, immunoblot was performed for the specific HAT, β-Actin loading control, and H3K14ac. (F-J). Each shRNA line was seeded at 2500 cells per well in a 24-well plate and treated with vehicle control or increasing doses of olaparib for eight days, after which colonies were fixed and stained with crystal violet. Stain was dissolved and the absorbance from each well was read by spectrophotometer, then normalized to vehicle control. IC50 curves were plotted in GraphPad Prism.
