Effect of GFP-EH21 on the endocytosis of the viral B5 protein. HeLa cells were infected with vEH21i in the absence or presence of IPTG. After 8 h, the cells were washed twice with ice-cold PBS and incubated with an anti-B5 MAb diluted in PBS for 1 h on ice. Cells were washed twice with PBS and either fixed immediately, permeabilized, and stained with Alexa 568-conjugated anti-rat IgG or incubated with fresh medium (with [+] or without [−] IPTG) for 1 at 37°C before fixing, permeabilizing, and staining.
