TGF-β signaling induces DUSP8 phosphatase activity. DUSP8 induces IL-9 transcription by dephosphorylating the transcriptional repressor Pur-α in the nucleus. Dephosphorylated Pur-α is exported from the nucleus to the cytoplasm in T cells, resulting in induction of IL-9 transcription. As a result, DUSP8 promotes Th9-mediated allergic diseases.
