a, Multiplexed sample preparation methods are shown the space of reaction volumes and number of cells that can be simultaneously prepared. Nanoliter reaction volumes increase efficiency of sample preparation by reducing adsorptive losses while reducing cost per cell. Slide based methods (nPOP) allow for the largest batch size, followed by microfabricated chip based methods (nanoPOTS41, N240, proteoCHIP12), and multiwell plate based methods, mPOP38,39. b, Time needed for the analysis of a single cell by 3-plexDIA (about 15 min active gradient and about 37 min total run time per labeled set) and by pSCoPE (about 60 min active gradient and 85 min total run time per labeled set).
