Notes: Polarized Caco-2 cells were treated with IAld (200 uM) for 1 h with or without 1 h prior treatment of CH-22 (10 μM), followed by LPS (100 ng/mL) stimulation for 4 h. (A) The levels of phosphorylated NF-κB p65 (p-p65) were determined by immunoblot. Total protein was a loading control. Data are presented as the ratio of p-p65/total p65. (B and C) Quantitation of p65 translocated ratio in Caco-2 cells and representative images of immunofluorescent staining of p65 translocation from cytoplasmic to nucleus. All data obtained from immunoblot or IF were quantified using Image J. The data are shown as the mean ± SEM (n = 3). *p < 0.05, **p < 0.01, ***p < 0.001.
