Table 4.

Effects of curcumin on As3+-induced mitochondrial membrane potential loss in rat hepatocytes

Incubation time Addition
9±2 5±3 2±1 Control rat hepatocytes
42±3a 21±4a 11±3a +As3+ (50 µM)
54±5b 29±3b 12±3 + Curcumin (10 µM)
36±5 19±4 6±3 + Curcumin (5 µM)
18±5b 7±3b 3±2b + Curcumin (2 µM)

Hepatocytes (106 cells/ml) were incubated in Krebs–Henseleit buffer pH 7.4 at 37◦C for 1.0 hr following the addition of EC50, 2hr of As3+. Mitochondrial membrane potential was determined as the difference in mitochondrial uptake of the rhodamine 123 between the control and treated hepatocytes and expressed as fluorescence intensity unit (10). Values are expressed as mean±SD of three separate experiments (n=3).a Significant difference in comparison with the control hepatocytes (p<0.05).b Significant difference in comparison with the As3+ treated hepatocytes (p<0.05).
