Effects of nptII cartridge insertions in the P. syringae pv. syringae 61 (Pss61) hrpF, hrpG, hrpT, and hrpV genes on the production and secretion of HrpZ. (A) Bacteria in early-logarithmic-phase growth in KB medium were harvested by centrifugation and resuspended at an OD₆₀₀ of 0.5 in Hrp minimal medium. After a further 5 h of incubation, the cultures were fractionated into cell (C) and supernatant (S) fractions by centrifugation and assayed for the accumulation of HrpZ and NptII by SDS-polyacrylamide gel electrophoresis followed by immunoblot analysis with appropriate antibodies and chemiluminescent immunodetection. (B) Bacteria were grown to stationary phase in KB medium and then handled as described above.
