A. Schematic of CRISPR screen. BMDMs, bone marrow-derived macrophages.

B. Lewis rat BMDMs were infected with indicated parasites (MOI = 1) for 24 h. Macrophage viability was measured via MTS assay. Data are displayed as paired plots for each individual screen.

C. Lewis rat BMDMs were infected with indicated parasites (MOI = 0.5) for 24 h. The number of parasites per vacuole was quantified by microscopy. A total of 100 to 120 vacuoles were counted per experiment. Data are displayed as mean + SD with two independent experiments. Significance was determined with two-way ANOVA with Tukey’s multiple comparisons test.

D. Top candidate genes with at least 3 enriched sgRNAs and significant enrichment (p-value < 0.05, analyzed by MAGeCK algorithm) after 4th round selection in both screens. Numbers between parentheses are the number of enriched sgRNAs after the 4th round of selection.
