Each vector in the Lenti-sgRNA/Cre vector library contains a sgRNA, a Cre, and a two-component barcode sgID-BC consisting of an 8-nt ‘sgID’ corresponding to each sgRNA and a random 15-nt barcode (BC). The Lenti-sgRNA/Cre is delivered to mice intratracheally. At appropriate times, entire lung of mice is digested, and DNA is extracted. Then, Tuba-seq is performed, followed by data analysis.
